Hormonal regulation of gluconeogenesis

The topic of this thesis is to point out which enzymes are affected by these hormones and to determine the molecular mechanism that underlies the activity changes. The involvement of cAMP and/or Ca 2 + in the phosphorylation of particular enzymes was investigated. A method was developed that could also be applied on human 1 iver biopsies. It is pointed out that a defect in the regulatory mechanism might lead to lactic acidosis. Furthermore the properties of Fru 1,6-P2 , an important allosteric effector of which the concentration changes rapidly under different hormonal conditions, and of the enzyme that catalyses the hydrolysis of this metabolite, FDPase, were investigated more closely. 2
